The original Great One, Jackie Gleason, would be my first choice. Ralph Kramden is one of television's most memorable characters, and the honeymooners is the greatest sitcom of all time, so much television comedy can be traced back to that show. There was a lot of pathos in Gleason's work, born of his troubled childhood. If you haven't seen it,check out a movie called "Soldier In The Rain" and you will see the full range of his talent.
John Goodman would be my number 2. He has played some very distinctive characters,particularly in the Coen brothers movies he has been in. Check out an underrated movie called "Matinée" that he was in, it's one of my favorite performances of his.
I agree with those who said that John Candy deserves to be higher up on the list. I think he has a lot in common with Gleason.
Kevin James should be on the list as well for both his stand up and his work as Doug Heffernan on "The King Of Queens".
My number one animated fat guy would be Homer Jay Simpson. D'oh pretty much says it all.
