The collision occurred in the southbound lanes of Highway 400 near Bass Pro Mills Drive around 3 a.m.
Police say a red Volkswagen Jetta was reportedly travelling at high speeds and rear-ended a minivan with four occupants inside, forcing the van to roll over.
A 73-year-old woman from North York was pronounced dead at the scene and three other occupants were taken to hospital with minor injuries.
The driver of the Jetta was last seen walking away from the scene. He is describe as being five-foot-10, between 35 to 40 years old with a thin build and braided hair. He was last seen wearing blue jeans and carrying a reusable shopping bag.
Police have appealed for him to “call a lawyer and turn themself in.”
